Mechanism of Action

Didanosine (ddI) is a nucleoside reverse transcriptase inhibitor (NRTI) that is phosphorylated intracellularly to ddATP. ddATP competes with natural dATP and causes chain termination in viral DNA due to the absence of a 3’OH group. It effectively blocks HIV reverse transcriptase and inhibits viral replication.

Used in HIV polymerase research, chaintermination modelling, NRTI SAR studies, antiviralresistance assays, and nucleosidemetabolism research.

Side Effects and Risks

Risks include pancreatitis, peripheral neuropathy, lactic acidosis, hepatic toxicity, GI intolerance and mitochondrial toxicity. Handle with antiviralnucleoside precautions.
