Mechanism of Action
Cladribine is a purine nucleoside analogue (2chlorodeoxyadenosine) that becomes phosphorylated intracellularly to its active triphosphate form. It incorporates into DNA and inhibits ribonucleotide reductase, leading to depletion of deoxynucleotide pools, faulty DNA synthesis and strand breaks. Lymphocytes are particularly sensitive due to low 5nucleotidase activity, causing selective depletion of B and Tcells.

Side Effects and Risks
