Mechanism of action
Betamethasone acetate is a slowly hydrolysed glucocorticoid ester that binds glucocorticoid receptors after enzymatic cleavage to active betamethasone. It suppresses proinflammatory gene transcription (e.g., NF-κB, AP1) and enhances antiinflammatory mediators. Micronisation increases surface area and enhances dissolution-rate characteristics.
Benefits and advantages
Used in corticosteroid pharmacokinetic modelling, depot-formulation research, inflammatory cytokine mapping, receptor binding studies and long-acting glucocorticoid formulations. Micronised forms are especially useful in dissolution studies and topical-delivery research.
Side effects and risks
Adverse effects include adrenal suppression, local irritation, dermal atrophy and systemic glucocorticoid effects with high exposure. Requires steroid-active containment handling.
