Mechanism of action
Beclomethasone is a glucocorticoid prodrug converted to its active metabolite beclomethasone17monopropionate (B17MP). The active form binds intracellular glucocorticoid receptors, modulating gene transcription by inhibiting NF-κB, AP1 and other proinflammatory pathways. It reduces cytokine release, leukocyte recruitment and mucosal oedema.

Used extensively in inflammation research, respiratory immunology, epithelial barrier models, corticosteroid receptor binding studies and inhalationpharmacokinetic simulations. Its welldefined receptor affinity and local activity support studies on topical vs systemic glucocorticoids.
Side effects and risks
Potential risks include mucosal irritation, adrenal suppression at high doses, dysphonia, oral candidiasis, and systemic glucocorticoid effects with prolonged exposure. Laboratory handling requires steroidactive containment.
