Mechanism of Action
Clozapine is an atypical antipsychotic with high affinity for D dopaminergic receptors and potent antagonism at 5HTA receptors. It also modulates histamine, muscarinic and adrenergic receptors. Clozapine reduces mesolimbic dopaminergic signalling while sparing nigrostriatal pathways, giving it strong antipsychotic effects with minimal extrapyramidal symptoms. It additionally enhances NMDAglutamatergic function through indirect modulation.
