Mechanism of Action
Clodronate disodium is a first‑generation non‑nitrogenous bisphosphonate that inhibits osteoclast‑mediated bone resorption. It is metabolized into non‑hydrolyzable ATP analogues inside osteoclasts, leading to apoptosis. It also reduces macrophage activity.
Benefits and Advantages
Used in bone‑resorption research, osteoclast apoptosis modelling, osteoporosis studies, skeletal‑metabolism assays and bisphosphonate SAR comparisons.
Side Effects and Risks
Risks include hypocalcemia, nephrotoxicity, jaw osteonecrosis (rare), GI irritation and flu‑like symptoms. Handle with bisphosphonate‑class precautions.
