Mechanism of Action
Cyclosporine is a cyclic undecapeptide immunosuppressant that binds cyclophilin, forming a complex that inhibits calcineurin phosphatase. This prevents dephosphorylation and nuclear translocation of NFAT (nuclear factor of activated T-cells), thereby blocking transcription of IL‑2 and other cytokines essential for T‑cell activation. It selectively suppresses cell‑mediated immunity while sparing myelopoiesis.

Used extensively in transplant‑immunology research, T‑cell signalling studies, calcineurin‑pathway modelling, autoimmune‑disease investigations, cytokine‑expression analysis, and drug‑interaction studies involving CYP3A4/P‑gp transport.

Risks include nephrotoxicity, hypertension, neurotoxicity, gingival hyperplasia, hirsutism, hepatotoxicity and immunosuppression‑related infection risk. Handle with full immunomodulator precautions.
