Mechanism of Action

Chlorproguanil is a dihydrofolate reductase (DHFR) inhibitor used in antimalarial research. It blocks protozoal DHFR, disrupting folate recycling and inhibiting thymidylate and purine synthesis required for DNA replication. This leads to cytostasis and death of rapidly replicating Plasmodium parasites.

Benefits and Advantages

Used in malaria-drug resistance studies, folate-cycle modelling, DHFR-mutation assays, combination-antimalarial research (e.g., with dapsone), and structure–activity investigations of antifolate compounds.

Side Effects and Risks

Risks include hepatotoxicity, GI disturbances, bone-marrow suppression, folate deficiency and hypersensitivity. Handle with antifolate safety protocols.
