Mechanism of action
Belimumab is a fully human monoclonal antibody targeting Blymphocyte stimulator (BLyS/BAFF), a cytokine essential for Bcell survival and maturation. By neutralising soluble BLyS, belimumab decreases survival signals for autoreactive Bcells, reducing autoantibody formation and modulating the adaptive immune response.
Benefits and advantages
Belimumab is crucial in autoimmunedisease research, particularly systemic lupus erythematosus models. It supports studies on Bcell tolerance, cytokinedriven autoimmunity, Bcell depletion strategies and interaction between BAFF signalling and germinalcentre dynamics.
Side effects and risks
Risks include infection, depression, infusion reactions, hypersensitivity and immunologic suppression. As a biologic, belimumab requires coldchain stability and biosafety handling.
