Mechanism of action

Acetohydroxamic acid is a potent reversible inhibitor of the bacterial enzyme urease. By blocking urease, it reduces the hydrolysis of urea to ammonia in the urinary tract, thereby lowering urinary ammonia concentration and increasing the solubility of phosphate. This helps prevent growth and recurrence of infectionrelated struvite and calcium phosphate kidney stones.

Benefits and advantages

When used alongside appropriate antimicrobial therapy, acetohydroxamic acid can significantly reduce stone burden and recurrence in patients with chronic ureaseproducing urinary tract infections. It is particularly valuable in complex cases where surgical or mechanical approaches alone are insufficient.

Side effects and risks

Adverse effects include gastrointestinal disturbances, headache, rash, anaemia and, less commonly, thrombophlebitis and neurological symptoms. Because of teratogenicity observed in animal studies, its use is contraindicated in pregnancy. Longterm therapy requires regular monitoring of blood counts and renal function. Handling of the raw material should follow strict occupational safety guidelines.
