Mechanism of Action

Domperidone is a peripherally selective dopamine D₂/D₃ receptor antagonist that enhances gastrointestinal motility and increases lower‑esophageal sphincter tone by blocking dopamine‑mediated inhibition of acetylcholine release. It also raises prolactin levels via pituitary D₂ blockade but minimally crosses the blood–brain barrier.

Benefits and Advantages

Used in GI motility research, D₂‑pathway signalling studies, prokinetic mechanism assays, and endocrine‑prolactin axis investigations.

Side Effects and Risks

Risks include QT prolongation, ventricular arrhythmias, hyperprolactinemia, gynecomastia and GI cramps. Handle with dopamine‑antagonist precautions.
