Mechanism of Action
Clozapine is an atypical antipsychotic with high affinity for D₄ dopaminergic receptors and potent antagonism at 5‑HT₂A receptors. It also modulates histamine, muscarinic and adrenergic receptors. Clozapine reduces mesolimbic dopaminergic signalling while sparing nigrostriatal pathways, giving it strong antipsychotic effects with minimal extrapyramidal symptoms. It additionally enhances NMDA‑glutamatergic function through indirect modulation.
