Mechanism of Action
Clodronate disodium is a firstgeneration nonnitrogenous bisphosphonate that inhibits osteoclastmediated bone resorption. It is metabolized into nonhydrolyzable ATP analogues inside osteoclasts, leading to apoptosis. It also reduces macrophage activity.
Benefits and Advantages
Used in boneresorption research, osteoclast apoptosis modelling, osteoporosis studies, skeletalmetabolism assays and bisphosphonate SAR comparisons.
Side Effects and Risks
Risks include hypocalcemia, nephrotoxicity, jaw osteonecrosis (rare), GI irritation and flulike symptoms. Handle with bisphosphonateclass precautions.
