Mechanism of Action
Cyclosporine is a cyclic undecapeptide immunosuppressant that binds cyclophilin, forming a complex that inhibits calcineurin phosphatase. This prevents dephosphorylation and nuclear translocation of NFAT (nuclear factor of activated T-cells), thereby blocking transcription of IL2 and other cytokines essential for Tcell activation. It selectively suppresses cellmediated immunity while sparing myelopoiesis.

Side Effects and Risks
Risks include nephrotoxicity, hypertension, neurotoxicity, gingival hyperplasia, hirsutism, hepatotoxicity and immunosuppressionrelated infection risk. Handle with full immunomodulator precautions.
