Mechanism of Action
Chlorthalidone is a thiazide-like diuretic that inhibits the Na⁺/Cl⁻ symporter (NCC) in the distal convoluted tubule. By reducing sodium and chloride reabsorption, it increases diuresis and lowers plasma volume. Unlike classical thiazides, its long half-life (40–60 hours) provides prolonged blood‑pressure control. It also enhances calcium reabsorption, lowers uric acid clearance and modulates vascular smooth‑muscle tone.

Used in hypertension-modelling studies, renal-electrolyte research, transporter-inhibition assays, long‑acting diuretic PK/PD modelling, nephrolithiasis studies and cardiovascular‑risk biomarker investigations.
Side Effects and Risks
Risks include hypokalemia, hyponatremia, hyperuricemia, hyperglycemia, hypotension and rare photosensitivity. Handle as a potent electrolyte-altering agent.
