Mechanism of Action

Chlorphenamine (chlorpheniramine) maleate is a first-generation H₁-antihistamine that competitively antagonises histamine at H₁ receptors. It reduces capillary permeability, vasodilation, pruritus and allergic inflammatory responses. Owing to its strong CNS penetration, it exhibits sedative, anticholinergic and antiemetic effects. It also shows weak serotonin-reuptake inhibition.

Used in allergy-modelling studies, histamine-receptor signalling research, antihistamine SAR evaluations, central-sedation pathway analysis, and behavioural pharmacology research. Frequently utilised as a reference sedating antihistamine.

Side Effects and Risks

Risks include sedation, cognitive impairment, anticholinergic effects (dry mouth, urinary retention, constipation), tachycardia, dizziness and paradoxical excitation in children. Handle as a CNS-active antihistamine.
