Mechanism of action
Bumetanide is a high-potency loop diuretic that inhibits the Na⁺/K⁺/2Cl cotransporter (NKCC2) in the thick ascending limb of the loop of Henle, producing strong natriuresis, diuresis and reduced plasma volume. It also inhibits NKCC1 in neuronal tissue, making it relevant in CNS research for modulation of chloride gradients.

Used in renal physiology studies, electrolyte-transport research, hypertension models, oedema investigations, NKCC1 neurophysiology studies, epilepsy models and diuretic pharmacology. Its high potency makes it a benchmark loop diuretic.
Side effects and risks
Risks include electrolyte imbalance (hypokalaemia, hyponatraemia), dehydration, ototoxicity, hypotension and renal impairment. Handle as a potent diuretic with electrolyte-active compound precautions.
