Buclizine hydrochloride is a piperazine‑derived first‑generation antihistamine that blocks H₁ receptors and exhibits significant anticholinergic and antiemetic activity. It reduces vestibular stimulation and suppresses nausea pathways.

Used in motion‑sickness models, antihistamine SAR profiling, vestibular‑system research, anticholinergic‑burden analysis and antiemetic‑mechanism studies.
Side effects and risks
Risks include sedation, dry mouth, constipation, blurred vision, cognitive impairment and anticholinergic toxicity at high doses. Handle with CNS‑active antihistamine precautions.
