Mechanism of action
Benperidol is a highly potent butyrophenone antipsychotic that acts as a strong antagonist at dopamine D₂ receptors, with additional interactions at D₃, D₄ and 5-HT₂ receptors. It suppresses mesolimbic dopaminergic transmission and reduces severe psychotic agitation.

Extensively used in dopaminergic research, receptor-binding studies, antipsychotic SAR mapping, hyperprolactinaemia models and CNS behavioural pharmacology. Its extreme potency makes it a key reference compound for high-affinity D₂ antagonism.
Side effects and risks
Risks include extrapyramidal symptoms, dystonia, akathisia, hyperprolactinaemia, QT prolongation and neuroleptic malignant syndrome. Handle with neuroactive and antipsychotic compound precautions.
