AMSACRINE
Amsacrine is an antineoplastic agent that intercalates into DNA and inhibits topoisomerase II, disrupting replication in rapidly dividing cells. It is used mainly in acute leukemias. Benefits include strong cytotoxic activity against resistant malignancies. Side effects may include myelosuppression, cardiotoxicity, nausea, vomiting, and mucosal irritation, requiring close monitoring during treatment. Mechanism of action
Amsacrine is an acridine-based topoisomerase II inhibitor that intercalates DNA, stabilising the cleavable complex and inducing double-strand breaks. This triggers apoptosis in rapidly proliferating cells.
Benefits and advantages
Widely used in oncopharmacology for studying DNA intercalation, topoisomerase inhibition, drugresistance mutations and apoptosis pathways.
Side effects and risks
Myelosuppression, cardiotoxicity, mucositis and nausea. Must be handled under cytotoxic containment.
