Mechanism of action
Ambroxol hydrochloride is a mucolytic and expectorant agent. It acts by depolymerising mucopolysaccharide fibres and altering the structure of bronchial mucus, thereby reducing viscosity and improving mucus transport. Ambroxol also stimulates the production and release of pulmonary surfactant and enhances ciliary activity in the respiratory epithelium.
Benefits and advantages
Through its combined effects on mucus rheology and mucociliary clearance, ambroxol facilitates expectoration in acute and chronic respiratory conditions associated with abnormal mucus secretion, such as bronchitis and COPD. Its local anaesthetic and antioxidant properties have additionally been explored for relief of sore throat symptoms. The compound has a long history of clinical use and is available in multiple oral and inhaled formulations.
Side effects and risks
Ambroxol is generally well tolerated, with the most common adverse events being mild gastrointestinal upset, nausea or dysgeusia. Rarely, serious cutaneous reactions such as Stevens–Johnson syndrome and toxic epidermal necrolysis have been reported, leading to cautionary labelling in some regions. As an API, ambroxol HCl should be handled with measures to limit dust and dermal exposure.
