Mechanism of action
Bendamustine hydrochloride is a bifunctional alkylating agent combining a nitrogen mustard moiety with a benzimidazole ring. It forms inter‑ and intrastrand DNA crosslinks, inducing apoptosis in rapidly proliferating cells. Bendamustine also triggers DNA‑damage responses, mitotic arrest and mitochondrial‑mediated cell death.

Crucial in cancer research including lymphoma, chronic lymphocytic leukaemia and solid‑tumour models. Used to study alkylator‑resistance mechanisms, DNA repair pathways, apoptosis signalling, and structure‑activity relationships among nitrogen mustards.

Risks include severe myelosuppression, nausea, mucositis, hepatotoxicity, infusion reactions and secondary malignancy risk. Requires full cytotoxic‑handling containment, closed systems and PPE.
