ACLACINOMYCIN A

Aclacinomycin A is an anthracycline antibiotic that inhibits DNA and RNA synthesis by intercalating into nucleic acids and disrupting polymerase activity. Mechanism of action

Aclacinomycin A (also known as aclarubicin) is an anthracycline antineoplastic antibiotic. It intercalates into DNA, inhibits topoisomerase II and generates free radicals, leading to disruption of DNA replication and repair and ultimately cell death, particularly in rapidly dividing cells.

Benefits and advantages

Aclacinomycin A has been used in the treatment of certain haematological malignancies, such as acute myeloid leukaemia, often in combination regimens. Compared with some other anthracyclines, it may exhibit a different toxicity profile, including potentially reduced cardiotoxicity at therapeutic doses.

Side effects and risks

As with other anthracyclines, dose‑limiting toxicities include myelosuppression, mucositis and alopecia. Cumulative cardiotoxicity with risk of congestive heart failure remains a concern, particularly at higher total doses or in susceptible patients. Extravasation can cause severe local tissue injury. Handling requires strict cytotoxic‑drug safety procedures.
