Mechanism of action
Alpha-ketoglutaric acid (2-oxoglutaric acid) is a central intermediate in the tricarboxylic acid (Krebs) cycle and plays a key role in cellular energy metabolism. It participates in oxidative decarboxylation reactions and serves as an important acceptor of amino groups in transamination pathways, thereby linking carbohydrate and amino acid metabolism. In nitrogen metabolism, alpha-ketoglutarate is a precursor of glutamate and glutamine and contributes to ammonia detoxification.
Benefits and advantages
Because of its pivotal metabolic position, alpha-ketoglutaric acid is used as a nutritional and functional ingredient in specialised medical nutrition, sports formulations and research media. It may help support mitochondrial function, muscle metabolism and nitrogen balance, particularly in catabolic states or intensive exercise. In combination with amino acids, it is used to formulate substrates that support tissue repair and recovery in clinical and performance settings.
Side effects and risks
At nutritional doses, alpha-ketoglutaric acid is generally well tolerated. Excessive intake, however, has the potential to influence acid–base balance and nitrogen handling, especially in individuals with impaired renal or hepatic function. As with all metabolic intermediates used in concentrated form, it should be formulated with appropriate attention to total dietary load. Industrial handling of the API requires standard measures to avoid dust formation, inhalation and contamination.
