Mechanism of Action

Doxepin hydrochloride is a tricyclic antidepressant with strong antagonism of H₁‑histamine receptors and moderate inhibition of norepinephrine and serotonin reuptake transporters. It stabilizes mood through monoamine elevation and produces profound antihistaminic sedation due to its very high H₁ affinity.

Benefits and Advantages

Used in depression‑modelling research, histamine‑receptor antagonism assays, sleep‑modulation studies, monoamine‑transporter SAR studies and chronic‑stress pathway modelling.

Side Effects and Risks

Risks include sedation, anticholinergic effects, weight gain, QT prolongation, arrhythmias and overdose toxicity. Handle with TCA‑class CNS‑active precautions.
