Mechanism of Action

Chlorothiazide is a thiazide diuretic that inhibits the Na⁺/Cl⁻ symporter (NCC) in the distal convoluted tubule. This reduces sodium and chloride reabsorption, increasing diuresis and decreasing extracellular fluid volume. It also reduces calcium excretion, providing secondary benefits in calcium‑stone research.

Used in renal‑physiology modelling, electrolyte‑transport studies, hypertension research, nephrolithiasis studies, NCC‑transporter inhibition assays and diuretic SAR investigations.

Side Effects and Risks

Risks include hypokalemia, hyponatremia, hyperuricemia, hypotension, glucose intolerance and rare photosensitivity. Handle as a potent electrolyte‑altering compound.
