Mechanism of Action
Chloroquine is a 4aminoquinoline compound that accumulates in parasite food vacuoles, raising vacuolar pH and inhibiting hemepolymerisation. Free heme becomes toxic, causing oxidative damage and parasite death. Chloroquine also modulates autophagy, lysosomal pH, and tolllike receptor signalling in mammalian cells.

Used in malariabiology research, lysosomalpH modulation studies, autophagy inhibition assays, antiviral pathway research, and tolllike receptor signalling investigations. A reference compound for endosomalacidification studies.
Side Effects and Risks
Risks include retinopathy, QT prolongation, neurotoxicity, hypoglycaemia, GI upset and dermatologic reactions. Handle as a high-potency lysosomotropic agent.
