Mechanism of action

Cefatrizine propionate is an orally bioavailable cephalosporin prodrug that is hydrolysed in vivo to the active antibiotic cefatrizine. The active moiety binds PBPs and inhibits cell-wall peptidoglycan crosslinking, leading to bactericidal effects against susceptible gram-positive and gram-negative organisms. The propionate ester enhances absorption and stability in the gastrointestinal tract.

Used in oral cephalosporin pharmacokinetics research, prodrug-conversion studies, β-lactam SAR profiling, gram-negative susceptibility assays and formulation-optimisation experiments. It is useful for modelling intestinal ester hydrolysis and systemic delivery of cephalosporins.

Side effects and risks

Risks include allergic reactions, GI disturbances, dysbiosis and rare hepatic or haematological abnormalities. Standard β-lactam handling precautions should be followed.
