Mechanism of action
Carbutamide is a first-generation sulfonylurea that stimulates insulin secretion from pancreatic β-cells. It binds to the sulfonylurea receptor 1 (SUR1) subunit of ATP-sensitive potassium (K_ATP) channels, causing channel closure, membrane depolarisation, opening of voltage-gated calcium channels and calcium influx. Elevated intracellular calcium triggers exocytosis of insulin granules.
