Mechanism of action

Buformin is a biguanide class antihyperglycaemic agent structurally related to metformin and phenformin. It reduces hepatic gluconeogenesis, increases peripheral glucose uptake and enhances insulin sensitivity. It also influences AMPK activation and mitochondrial respiratory-chain function.

Benefits and advantages

Used in metabolic-disease research, AMPK-signalling studies, gluconeogenesis modelling, biguanide SAR analysis, mitochondrial function studies and diabetes drug-development. Its potency and distinct PK profile make it useful in comparisons among biguanides.

Side effects and risks

Risks include lactic acidosis (especially in renal impairment), GI upset, fatigue and metabolic acidosis. Handle under biguanide safety protocols with strict dose control.
