Mechanism of action
Benzthiazide is a thiazide diuretic that inhibits the Na⁺/Cl⁻ cotransporter (NCC) in the distal convoluted tubule, increasing urinary sodium and chloride excretion. This reduces extracellular fluid volume and lowers blood pressure. Benzthiazide also decreases urinary calcium excretion and exhibits a modest vasodilatory effect through chronic reduction of intracellular sodium in vascular smooth muscle.

Used in renal-transport studies, distal-tubule physiology models, antihypertensive pharmacology, electrolyte-regulation investigation and comparative studies of thiazide diuretics. Also useful for analysing NCC binding behaviour and compensatory RAAS activation.
Side effects and risks
Risks include hypokalaemia, hyponatraemia, hyperuricaemia, glucose intolerance, photosensitivity and hypotension. Handle under diuretic- active safety standards with attention to electrolyte-active dust.
