Mechanism of action
Atenolol is a cardioselective -adrenergic receptor antagonist. It preferentially blocks receptors in cardiac tissue, leading to decreased heart rate (negative chronotropy), reduced myocardial contractility (negative inotropy) and reduced renin secretion from juxtaglomerular cells. These effects result in lowered blood pressure and diminished myocardial oxygen demand, with reduced bronchial blockade compared with non-selective beta-blockers.
Benefits and advantages
In cardiovascular research, atenolol is a standard reference compound for exploring -selective blockade, baroreflex physiology, sympathetic modulation and comparative beta-blocker pharmacodynamics. It is also useful in modelling reninangiotensin system interactions and heart-rate control in exercise or stress protocols.
Side effects and risks
Risks include bradycardia, atrioventricular conduction disturbances, hypotension, fatigue, cold extremities and, in susceptible patients, worsening heart failure. Abrupt withdrawal can provoke rebound tachycardia or angina. Laboratory handling should minimise dust generation and exposure due to its potent cardiovascular activity.
