Mechanism of action
Amodiaquine is a 4‑aminoquinoline antimalarial with a mechanism similar to chloroquine but with efficacy against chloroquine-resistant Plasmodium falciparum strains. Within the parasite food vacuole, amodiaquine interferes with haem detoxification, preventing polymerisation of haem into inert haemozoin. Accumulated free haem exerts oxidative and membrane toxicity, resulting in parasite death.
Benefits and advantages
Widely used in malaria pharmacology and pharmacoresistance research, especially in modelling ACT (artemisinin combination therapy) efficacy. Its metabolic formation of desethylamodiaquine provides a clinically relevant active metabolite for PK studies.
Side effects and risks
Rare but potentially severe hepatotoxicity, agranulocytosis and neurotoxicity have been reported. Handling requires strict contamination control.
