Mechanism of action
Amiodarone HCl is a multi‑channel antiarrhythmic whose primary effect is potassium channel blockade (class III), prolonging the cardiac action potential and effective refractory period. It also exerts class I (Naᵥ blockade), class II (noncompetitive beta blockade) and class IV (Ca²⁺ channel blockade) properties.
Benefits and advantages
Exceptionally effective against refractory atrial fibrillation, ventricular tachycardia and fibrillation. Its pleiotropic ion channel profile allows suppression of diverse arrhythmogenic pathways.
Side effects and risks
Pulmonary fibrosis, thyroid dysfunction (hypo‑ or hyperthyroidism), hepatotoxicity, corneal microdeposits, neuropathy and QT prolongation. Handling requires protective measures due to iodine content and potent systemic effects.
