Mechanism of action
Amezinium methylsulfate is a sympathomimetic agent that increases arterial blood pressure by enhancing noradrenergic transmission. It acts mainly by inhibiting neuronal reuptake of noradrenaline and by promoting its release from sympathetic nerve endings, leading to increased activation of α-adrenergic receptors in vascular smooth muscle and consequent vasoconstriction.
Benefits and advantages
Amezinium has been used in the management of certain forms of hypotension, such as orthostatic hypotension, where elevation of systemic vascular resistance can alleviate symptoms like dizziness and syncope. Its pharmacodynamic profile allows oral dosing with relatively predictable haemodynamic effects under supervised conditions.
Side effects and risks
Adverse effects reflect enhanced sympathetic activity and include headache, palpitations, tachycardia, piloerection, urinary retention and, in some cases, supine hypertension. It should be used cautiously in patients with cardiovascular disease, arrhythmias or urinary outflow obstruction. As an API, amezinium methylsulfate requires careful handling to prevent accidental exposure and should be stored securely.
