Mechanism of action
Amantadine sulfate is the sulfate salt of amantadine, providing improved stability and solubility for certain formulations. It shares the same antiviral and antiparkinsonian mechanisms as the hydrochloride form, blocking the M2 proton channel of influenza A virus and modulating NMDA receptor-mediated neurotransmission and dopamine release in the central nervous system.

The sulfate salt offers formulators an alternative option for solid or liquid dosage forms, with physicochemical properties that may be advantageous in specific product designs. It serves as a functionally equivalent alternative where regulatory or technical considerations favour the sulfate over the hydrochloride salt.
Side effects and risks
Side effects are essentially the same as those of amantadine hydrochloride, including central nervous system effects (insomnia, dizziness, confusion), anticholinergic-like symptoms and peripheral oedema. Careful titration, patient selection and renal dose adjustment remain important. Manufacturing and handling must follow standard precautions for centrally active agents.
