ACEMETACIN

Acemetacin is an NSAID that inhibits cyclooxygenase enzymes, reducing prostaglandin synthesis and providing analgesic and anti-inflammatory effects. It is used for arthritis and pain management. Mechanism of action

Acemetacin is a non‑steroidal anti‑inflammatory drug (NSAID) and prodrug of indometacin. After administration it is rapidly de‑esterified to indometacin, which non‑selectively inhibits cyclo‑oxygenase (COX‑1 and COX‑2) and reduces the synthesis of prostaglandins involved in pain, inflammation and fever.

Benefits and advantages

Compared with indometacin itself, acemetacin may offer improved gastrointestinal tolerability while maintaining anti‑inflammatory and analgesic efficacy. It is used in rheumatic diseases, musculoskeletal disorders and other painful inflammatory conditions when formulated in appropriate dosage forms.

Side effects and risks

As with all NSAIDs, the main risks involve gastrointestinal irritation, ulceration and bleeding, renal impairment, fluid retention and potential cardiovascular events with long‑term use. Hypersensitivity reactions, including bronchospasm in aspirin‑sensitive individuals, can occur. The API should be handled with care to avoid dust inhalation or skin contact, and finished medicinal products require careful patient selection and monitoring.
