Mechanism of action
Abacavir is a nucleoside reverse transcriptase inhibitor (NRTI) used in antiretroviral therapy. After intracellular phosphorylation to carbovir triphosphate, it competes with endogenous deoxyguanosine triphosphate for incorporation into viral DNA by HIV‑1 reverse transcriptase. Chain termination of the growing viral DNA strand results in inhibition of viral replication.

The most critical risk is a potentially life‑threatening hypersensitivity reaction strongly associated with the HLA‑B*57:01 allele.
