Mechanism of Action
Cinacalcet hydrochloride is a calcimimetic agent that allosterically activates the calcium‑sensing receptor (CaSR) on parathyroid chief cells. By increasing the receptor’s sensitivity to extracellular calcium, it suppresses PTH secretion, modulates downstream calcium–phosphate homeostasis, and influences bone‑remodelling pathways. It does not act as an agonist itself but enhances the receptor’s responsiveness to physiological ligands.

Used in CaSR‑signalling research, hyperparathyroidism‑modelling studies, mineral‑metabolism analyses, bone‑remodelling research, and endocrine‑regulation modelling. Valuable for evaluating CaSR mutations and receptor allosteric modulation profiles.

Risks include hypocalcemia, QT prolongation, seizures (via calcium shifts), GI upset and myalgia. Handle as a potent endocrine‑modulating compound.
