Mechanism of action
Anastrozole is a selective, nonsteroidal aromatase inhibitor that blocks the conversion of androgens (testosterone, androstenedione) into estrogens via inhibition of the cytochrome P450 enzyme CYP19A1. By reducing circulating estradiol levels, anastrozole strongly suppresses estrogendependent signaling in breast tissue and other estrogenresponsive organs. Its mechanism involves reversible competitive inhibition at the aromatase complex in peripheral tissues, especially adipose tissue, liver, and tumor cells.
