Mechanism of action
Amorolfine is a morpholine antifungal agent that inhibits two key enzymes in ergosterol biosynthesis: Δ‑14-reductase and Δ‑7–Δ‑8-isomerase. Blocking these enzymes results in accumulation of abnormal non-ergosterol sterols, causing structural defects in fungal cell membranes and eventual fungal cell death.
Benefits and advantages
Amorolfine exhibits broad-spectrum activity against dermatophytes, yeasts and moulds, making it valuable in onychomycosis and dermatophyte pharmacology research. Its ability to penetrate keratin structures is particularly useful in permeability studies.
Side effects and risks
Mostly limited to local irritation or erythema. Systemic absorption is minimal but laboratory precautions against dermal exposure remain essential.
