Mechanism of action
Amlodipine is a dihydropyridine-type L‑type calcium channel blocker that preferentially inhibits voltage-dependent Ca²⁺ influx in arterial vascular smooth muscle. The reduction in intracellular calcium prevents actin–myosin cross-bridge formation, resulting in vasodilation and reduced systemic vascular resistance. Due to slow receptor association/dissociation kinetics, amlodipine produces sustained 24‑hour antihypertensive effects with minimal reflex tachycardia.

Side effects and risks
Peripheral oedema, flushing, fatigue and gingival hyperplasia are common. Rare but serious reactions include hypotension and hepatic enzyme elevation. Proper PPE is required due to vasodilatory potency.
