Mechanism of action
Alverine citrate is an antispasmodic agent that acts directly on smooth muscle, particularly in the gastrointestinal tract and uterus. Although its precise molecular targets are not fully defined, it appears to reduce smooth muscle contractility by modulating calcium influx and intracellular calcium handling without significant anticholinergic activity.
Benefits and advantages
Alverine is used for the symptomatic relief of abdominal pain and cramps associated with irritable bowel syndrome and other functional bowel disorders. It may also be employed in dysmenorrhoea. Because it lacks strong anticholinergic or opioid effects, it tends to have a relatively favourable tolerability profile and does not impair normal gastrointestinal motility when used appropriately.
Side effects and risks
Most adverse effects are mild and may include nausea, dizziness, headache or transient gastrointestinal discomfort. Hypersensitivity reactions, including rash or, very rarely, severe allergic responses, have been reported. As with all antispasmodics, use should be individualised based on patient characteristics. Handling of the API should minimise dust inhalation and direct contact.
